Season 2
Producer Paul Scheuring: Some characters will die, new people will be slid into place. There will be a lot more revelations about people’s pasts once they are outside the prison walls. I’m not going to say in what episode the actual break will happen, but the whole show will be reinvented. It’ll be ‘Planes, Trains and Automobiles,’ it’ll be every method of conveyance, as everyone runs around the country. It’ll be ‘The Great Escape.’ Ultimately it becomes about all of them fulfilling their individual destinies, with the whole of America on their [trails]. It’ll be ‘The Fugitive’ times six or 10. link
During the "Behind the Walls" special, Wentworth Miller mentioned that the story continues after the prison break by following the lives of the escapees, including Sucre and T-Bag, so there's no suspense surrounding those two being killed off early. link

Next Seasons
Writer-producer Nick Santora: We know where the show's going through Season 4, and we have ideas through Season 5. We spent the first few days after Thanksgiving breaking out details for the end of [several] episodes of Season 2 and doing the story arcs for Season 3 in great detail. We know exactly where it's going.
Producer Paul Scheuring: There is a larger canvas here, and it is longer than a season," he says. "It's not going to be something like '24,' where we're perpetually reinventing a new obstacle to overcome each season. Season Two will just be the escalation of Season One.
